Breast cancer (BC), a prevalent malignant disease, is frequently observed among women worldwide. For better prognoses, it is vital to seek advancements in diagnostic and treatment procedures. Research on protein kinases, including PKMYT1, a membrane-associated tyrosine/threonine kinase from the Wee family, has been conducted in some tumor types, not including breast cancer (BC). The functional role of PKMYT1 was investigated in this study, combining bioinformatics methods with the analysis of local clinical samples and experimental procedures. The comprehensive study showed that PKMYT1 expression was increased in breast cancer tissue, more apparent in individuals with advanced disease, in contrast to the levels observed in normal breast tissue. For breast cancer patients, PKMYT1 expression levels were an independent factor influencing prognosis when considered with their clinical characteristics. Following a multi-omics investigation, we determined a close association between PKMYT1 expression levels and several oncogenic or tumor suppressor gene mutations.
